The Romanesque Revival red brick courthouse was built in 1903. It is located on Walnut Street and Brunswick Street. United States Highway 341 is two blocks north of the site. The building was enlarged in 1975 and renovated in 1999. One source has the architect to be James Wingfield Golucke while another says S. A. Baker provided the designs. The building features a tower with a working clock. Veteran memorials are on the grounds. Wayne County was formed on December 7, 1803 from Creek Lands as the 29th county. Tuckersville (1801), Waynesville (1829) and Jesup (1873) have both served as county seat. The county is named for Revolutionary War General Anthony Wayne. The county seat is named for General Thomas Sidney Jesup. The county center is 7.6 miles South-Southwest of Jesup.

Twenty-one Superior Court Judges serve Wayne, Appling, Bacon, Brantley, Bryan, Bulloch, Camden, Charlton, Chatham, Coffee, Effingham, Evans, Glynn, Jeff Davis, Jenkins, Liberty, Long, McIntosh, Pierce, Screven, Tattnall, and Ware counties and a State Judge and Four County Judges, court clerk, and county attorney serve Wayne County (judicial.) The county is located in southeast Georgia. Savannah, Georgia is northeast and Valdosta, Georgia is southwest of the county. The Altamaha River forms its northeast border. The county is surrounded clockwise by Tattnall, Long, McIntosh, Glynn, Brantley, Pierce, and Appling counties.
The area of the county is 645 square miles. It is 10 out of 159 counties in the state. It ranks 59 out of 159 in population in the state. It has a density of 46.7 persons per square mile making it 95 out of 159 in the state. Wayne County has 36.7% of its population in its incorporated areas. United States Highway 25 enters the county from the northeast, Long County, and exits to the southeast, Glynn County. United States Highway 84 goes northeast to southwest originally joined with United States Highway 25 from Long County, splits, and goes into Pierce County. United States Highway 301 is also joined with United States Highway 25 from Long County, splits, and heads south into Brantley County. United States Highway 341 enters from the northwest, Appling County and joins United States Highway 25 to travels southeast into Glynn County. The county is mostly triangular with the southwest corner removed. Jesup is located in the northern third of the county. Jesup is the county seat and the largest city. It is 32.6% of the county population. This county is in the Jesup Micropolitan Statistical Area.
